Especially the bacteriocins produced by lactic acid bacteria has been regarded as a safer food supplements since lactic acid bacteria is food grade bacterium. Resent years, some studies indicated that some antimicrobial peptides have been discovered with anticancer activity.
Our laboratory has been working on antimicrobial peptides that produced by lactic acid bacteria. In this research, four potential antimicrobial peptides had been selected from Lactobacillus casei group database. The expression vectors pET32a(+) with target genes have been constructed and were expressed in E. coli expression system. Four peptides purified by affinity column chromatography show different solubility with different E. coli strains and induction temperature. Origami2 produced more soluble particle among all the peptides. DII and EII can express at 37°C, while DI and EI express at 16°C. There is no antimicrobial activity among four peptides with test strains, but anticancer activity has been observed. Four peptides can cause cell death of SW480 (human colon adenocarcinoma) with different degree, and have less effect on H183B5F5/M10 (human mammary epithelial cell). It is no doubt about that revealing the mechanism of these peptides has benefit in human beings.
