The purpose of this study was to find optimum extraction conditions for antioxidants in black soybean seed coat and pre-baked black soybean and then to evaluate the regulatory effect of the extracts on blood lipids using high-fat diets induced hamster as the animal model. Black soybean seed coat and pre-baked black soybean (at 130℃ for 3 min) were extreacted either by ethanol (80%, 40%) or acetone (60%, 20%) at pH3 or pH5 and temperature of 4 or 30℃ for 24 h and then were vacuum concentrated and freeze-dried to measure the antioxidants content and antioxidant capacity. The results showed that pre-baked black soybean had high tannin content, total anthocyanin, iron chelating ability and reducing power when it was extracted using 60% acetone (pH 5) at 4℃. Under the same extraction condition, black soybean seed coat also obtained high tannins, total polyphenolic content, ABTS scavenging and reducing power. The variables in response surface methodology (RSM) were acetone concentration (40%, 60%, 80%), pH (4, 5, 6) and extraction time (1, 2, 3 days). The results showed that 80% acetone (pH 5) extraction at 4℃ for 36 h could achieve the maximum total anthocyanins content (12.8 mg/g). The effect of the extract on blood and fecal lipids contents was evaluated using Hamster as animal model. Hamsters were separated into four experimental groups : control group (AIN-93 diets group), high fat diet group (containing 40% fat, 0.2% cholesterol), high fat diet and the extract from black soybean seed coat at low dose (200 mg/kg) or high dose (800 mg/kg). During 12 weeks experiment, the extract of black soybean seed coat was orally administrated once daily. The model goup showed increased serum triglycerides, total cholesterol, LDL, aspartate aminotransferase and alanine aminotransferase, indicating dyslipidemia induced by high-fat diets. However, the extract could significantly reduced serum total cholesterol, triglycerides and LDL in high fat treated hamster; especially the reduction of 20% total cholesterol, 52% triglyceride and 44% LDL was found at high dose. The results of this study prove that anthocyanin-enriched extract from black soybean seed coat has the regulatory effect on serum blood lipids, thus, black soybean has the potential as the source for health improving foods. 許成光 學位論文 ; thesis 123 zh-TW |
