The use of anti-clotting substances may effectively reduce reduce thrombosis, and prevent the occurrence of cardiovascular disease. Vitis thunbergii var. taiwaniana, a conventional food and medical plant in Taiwan, has been reportedly to possess the bioactivities of anti-oxidation, anti-inflammation, anti-cancer, and anti-coagulation. The aim of this research was to analyze and identify the compounds with anti-coagulative activity in V. thunbergii var. taiwaniana. The roots were extracted with of 80% methanol and then concentrated under vacuum. The fraction was suspended by isopropanol, followed by the addition of salt to drive certain compounds to isopropanol layer. Separation in open column chromatography and HPLC were performed to isolate and purify anti-coagulants of isopropanol layer with the aid of bioassay-guided fractionation based on the time duration of intrinsic-coagulation and extrinsic-coagulation. NMR analysis revealed four potential anti-coagulants, (+)-catechin, amurensin G, hopeaphenol, (+)-viniferol D. Among which, amurensin G and (+)-viniferol D are the first time isolated from V. thunbergii var. taiwaniana. Collectively, the V. thunbergii var. taiwaniana may have several anti-coagulants with distinct anti-coagulaive mechanisms.
