The differentiation of chick embryo myoblasts in vitro was studied using electron paramagnetic resonance (EPR) and dielectric relaxation. Cell membranes of myoblasts in rotation culture for 72 hours were examined with three nitroxystearate (NSA) spin labels which insert at increasingly greater depths into the lipid bilayer (5-NSA < 12-NSA < 16-NSA). Changes in membrane order during myogenic differentiation in vitro were seen. The spin label which inserts near the bilayer surface (5-NSA) showed two peaks of membrane order: one at 15 hours (associated with gyratory aggregation and recovery of the myoblasts from dissociation) and one at 38 hours (associated with prostaglandin binding and increased cell ...
