Description
Summary:First this thesis outlines the historical background and current status of diabetes therapeutics. This includes prolonged action preparations, oral hypoglycaemic agents, approaches to peroral insulin therapy and methods of insulin modification, with emphasis on derivatives of clinical interest. Then the analytical methods are described. These include a novel method, using chromatography on silica-gel loaded paper, for analysing the state of acylation of water-insoluble insulin derivites. The next two chapters describe the synthesis of the acyl hydroxysuccinimidyl esters and the protected insulin derivites, followed by the coupling, purification and analysis of the products.
