Summary:Myocardial infarction increases endogenous anandamide release which may protect against ischaemia and reperfusion induced arrhythmias via an effect on coronary artery reactivity. The vasorelaxant of anandamide has been observed in many isolated vascular preparations but the mechanism remains unclear. Sphingosine-l-phosphate (SIP) has been implicated in the response as cannabinoid and SIP receptors have a high degree of structural similarity. Hence the aims of this study were to delineate the mechanism of action of anandamide and SIP in the rat coronary artery, to identify whether anandamide or the anandamide breakdown inhibitor IIRB597 could exert protective effects on ischaemia and reperfusion induced arrhythmias and to measure plasma levels of anandamide.
