This microscope has advantages of common path configuration and free of speckle noise that is not only to immured environment vibration, but also with the ability to enhance image contrast In this microscope configuration, using SAVART prisms as the shear plate and liquid crystal as phase-shifting device, The SAVART prism separates the incident beam into two parallel beams, which vibrate perpendicular to each other, To extract the phase gradient from the interference image ,The liquid crystal phase-shifting device is applied by input difference voltage to produce a biased phase shift of π/2 for each step , The different shearing direction image can be obtained by rotating the angle of the SAVART prisms. We also test two transparent samples ,which include human cheek cell and 25um period quartz transmission grating. The experiments and experimental results demonstrate the validity and ability of the transmission SAVART shearing interferometer microscope with liquid crystal phase-shifting method.
