The high concentration of uric acid in blood could make Monosodium urate crystals (MSU) accumulated in joints and different tissues, and thus induce Gout and Gouty arthritis. Plectranthus amboinicus (Lour.) Spreng is a perennial herb belonging to the family Lamiaceae which has been widely distributed in Taiwan. In this study, P. amboinicus (Lour.) Spreng leaf (PASL) extracted by three different solvent as distilled water, 50% ethanol and 95% ethanol. The extracts were subjected to analyses including total polyphenol content, 1,1-diphenyl-2-pycrylhydrazyl (DPPH) scavenging activity, xanthine oxidase inhibition activity, carvacrol and thymol contents. In addition, the optimized fertilizing method for P. amboinicus (Lour.) Spreng were also investigated according to 3414 fertilizing test (14 formulas) based on content of indicator component. As the result, the polyphenolic content in 95% ethanol extract was higher than that with that in aqueous extract and 50% ethanol extract (p<0.05). Similar results were also found in DPPH scavenging activity, xanthine oxidase inhibitory activity and carvacrol analysis. Thymol content is lower than the limit of quantitation (0.05 ppm) in all extracts. Carvacrol was thus the major indicator component in P. amboinicus (Lour.) Spreng. In case of cultivation test, nitrogen and phosphate in fertilizer were helpful on the yield and indicator component for cultivation of P. amboinicus (Lour.) Spreng, but excessive potassium in fertilizer inhibited its growth. In conclusion, 95% ethanol is the optimum extraction solvent for the PASL, and the fertilization conditions is 270(N), 90(P2O5) and 200(K2O) mg/kg. The results would provide important information on developing health food or alternative drug for Gout disease, and plant cultivation condition in the future.
