Sleep disorders or insufficient sleep can lead to chronic, systemic low-grade inflammation. Chronic inflammation over a long period of time can cause a variety of related diseases with inflammatory components, such as diabetes, atherosclerosis and neurodegeneration. Sleep disorders are mainly treated with antidepressants, melatonin and low doses of antipsychotic drugs. Ganoderma lucidum polysaccharides have been studied for the treatment of many diseases, such as sedatives, restlessness, insomnia and palpitations. It has been shown to protect nerves and improve anti-inflammatory response. We studied whether Ganoderma lucidum mycelium by solid - state fermentation could alleviate the damage of Mouse Neuroblastoma Cell (N2A) and Rat normal small intestine Cell (IEC-6) and improve the sleep function of mice by two extraction methods. In this study, LPS was used to induce inflammation in cells, and the expression of RNA, ELISA and Western Blot was used to evaluate the inflammation in vitro. In vivo experiments, irregular sleep-wake patterns caused by persistent and long-term light exposure in animals resulted in sleep disorder/depression patterns. Hypnosis-enhancement, Elevated-Plus Maze, Open Field Test and Pathological Section were used to see whether Ganoderma lucidum mycelium by solid - state fermentation could increase the sleep function of mice.The results showed that N2A and IEC-6 could effectively inhibit the expression of IL-1beta, TNF-alpha, NF-kappa B, iNOS and COX-2 in the mRNA. Hypnosis-enhanced test showed that mice fed Ganoderma lucidum solid - state fermentation could slow down barbiturate tolerance and prolong sleep duration.In the protein expression and pathological section color of the forehead, we can see that the expression of BDNF in the brain given Ganoderma lucidum mycelium by solid - state fermentation has been improved.
