Summary: | 碩士 === 國防醫學院 === 牙醫科學研究所 === 107 === Background: Periodontitis is an inflammatory disease caused by the aggregation of specific periodontal disease-related bacteria on the surface of the root, and the degree of periodontium damage depends on the host immune response. The immune system can be divided into innate immunity and adaptive immunity, and the complement system represents a major part of the innate immunity. Many diseases have been reported to be related to the disregulation of the complement system, and the relationship between periodontitis and the complement system has been discussed in many articles. At present,the diagnosis of periodontitis is based on clinical parameters. There is still a lack of discriminative biomarker to help diagnosis of periodontitis. Complement proteins C3b and C4b are central products of the complement system activation, however, research regarding the relationship between C3b and C4b and periodontitis is lacking and whether C3b and C4b could potentialy help clinicians diagnose periodontitis is unknown.
Aim: The purpose of this study was to investigate the relationship between complement protein C3b and C4b and periodontitis and their potential application for clinical diagnosis.
Materials and Methods: Samples from different animal models and gingiva sample/gingival crevicular fluid form patients with periodontitis were used to examine whether the expression levels of C3b and C4b were different between healthy and disease, and to investigate the correlation between the expression levels of C3b and C4b and disease severity. In addition, the opitimal cut points for diagnosis were calculated and selected by using SPSS.
Results: Our data show that the expression of C3b and C4b increased of samples in diseased sites from different animal models and from human studies. The expression of C3b and C4b may have positive correlation withthe severity of disease. For diagnostic development, C3b and C4b may have potential for diagnosis of periodontitis.
Conclusion: It can be seen that the complement proteins C3b and C4b may play an important role in the process of periodontal disease and may have the potential to become a tool for diagnosing periodontitis.
