For cancer treatment with chemotherapy, serious side effects are caused by the inability of the drug to be fully delivered to the tumor. Although many studies have pointed out that ultrasound can enhance drug delivery, it has not achieved the goal of majority drugs molecules to accumulate onto or around tumors. In order to reduce side effects and enhance the efficient killing of cancer cells, this study mainly uses a special targeting aptamer (AP52) and anticancer drug (Doxorubicin) conjugate to couple with ultrasound for cancer treatment. We also compare the results for both cancer and normal cell lines to explore the targeting effect of aptamer drugs. Methods: Breast cancer cells (MCF-7) and breast cells (MCF-10A) were used for the experiment. The ultrasonic frequency was 1 MHz, the intensity was 0.8 W/cm2, and the ultrasound irradiation duration was 60 seconds. Conclusion: The combination of doxorubicin-containing aptamer and ultrasound can enhance the drug delivery and efficiently kill cancer cells with less side effects. Significance: Aptamer conjugated to an anticancer drug can be used for targeting cancer cells and ultrasound can enhance drug delivery.
