Summary: | 碩士 === 國立交通大學 === 光電工程研究所 === 107 === This thesis study applied Fourier-transform visible-near infrared spectroscopy to analyze diffuse reflectance from human skin. We designed three effective methods to perturb the optical properties of skin tissues and were able to measure differential spectra which are about hundred times weaker than that of the original diffuse reflectance signal. We then employed the principal component analysis (PCA) method to retrieve three perturbation-induced characteristic spectra of human skin, which are attributed to be originated from the Rayleigh scattering and optical absorption by hemoglobin and melanin. We then developed an in vivo optical model of skin and applied Monte Carlo for Multilayered Media (MCML) algorithm to simulate the diffuse reflectance spectrum of skin tissue. The simulated spectra agree well with our measured data. Finally, we applied the spectral technique to quantify the physiological status of skin in vivo and monitor the optical changes of skin after mechanical impact. Our spectral technique can reveal the influences of hot and cold compresses on skin tissues after mechanical impact.
