Amphetamines appear in urine within three hours and are detectable after abuse. In this study, citrate stabilized gold nanoparticles were functionalized with self-assembled monolayers (SAMs) thioctic acid and were characterized with DLS, FT-IR, TEM, SEM/EDX and etc. AuNPs acts as not only the support but also the mass barcode for the determination of amphetamines with LDI-TOF MS analysis. First, AuNPs were functionalized with O-(2-Carboxyethyl)-O-(2-mercaptoethyl) heptaethylene glycol (SH-CH2-CH2-PEG8-COOH). Subsequently, the N terminus or primary amine of lysine residues on antibody were conjugated with activated carboxylic group by EDC/ Suflo-NHS, that could be used as a probe for the determination of targeted antigens. Mass spectrometry imaging (MSI) is a microscopy technique which could directly analyze hair without complex sample preparation. Brief summary, the development of methods possessed these advantages, including simple for fabrication, low-cost, high efficacy of separation, reproducibility and rapid determination for the targeted analytes. We expected this technique could be extensively applied not only for the forensic science but also for the diagnosis of diseases which possess specific antibodies to recognize disease related biomarkers.
