Summary: | 碩士 === 國立中興大學 === 化學系所 === 107 === It is accustomed in society to refer to abused drugs as the use of recreational drugs and other intoxicating substances, that can induce dependency (i.e. addiction) and cause abstinence. Emerging drugs of abuse are an important concern for the health as well as law enforcement. In addition to overdose leading to death, there are complications, such as brain degeneration, damage of the nervous system and harm of organs.
In this study, a sample and fast method for the preparation of antibody decorated functional gold nanoparticels (AuNPs) was developed for the determination of abused drug in hair. As the preliminary rapid screening of drugs with immunoassay shows a positive result, the laser desorption/ionization time of flight mass spectrometry (LDI-TOF MS) can be further applied to quantify the drug content in the hair sample, and the subject is judged to be in violation of the law according to the regulations of the local country. Traditional methods for the measurement of abused drugs took several hours to several days from biological fluids (serum, urine or saliva). Hair assay can detect long-term the use of abused drugs. Using mass spectrometry imaging, the complete distribution of the drug in the hair could be obtained.
Using the synthesized AuNPs probe to enhance the MS signal intensity and the probes presented high specificity under LDI MS analysis from the complex biological samples. According to the results, it can be easily discerned 3,4-Methylenedioxymethamphetamine (MDMA) in the hair, via the use of spiked hair with standard to obtain mass spectrometry image, and the detecting range low to 1 ng/mL.
