According to the World Health Organization guidelines, it can be classified as adenocarcinoma, signet ring-cell carcinoma, and undifferentiated carcinoma. Adenocarcinoma is the most common gastric cancer, accounting for 90~95% of gastric cancer patients. It has been found that the gastric cancer has several risk factors, such as heredity and diet, the infection of Helicobacter pylori is also an important factor in development of gastric cancer, long-term infection with Helicobacter pylori leads to chronic inflammation of gastric mucosa, which can cause the atrophic gastritis and even the pre-cancerous lesions of the mucosa. Previous studies indicated high expression of Helios is a significant better prognostic factor in gastric cancer patients, especially in H. pylori¬-infected and advanced gastric cancer patients. The aim of this research is to investigate the role of Helios in gastric tumor. The Helios is encoded by the gene IKZF2, Using the CRISPR/Cas9 system, we found that the growth rate of cells does not affect in cell experiment, but rather increases the ability of cells to migrate and invade. In vivo, the subcutaneous injection of Helios knockout gastric tumor cells showed that the tumor appeared to be slower than the parental cell line. In addition, it was also given intravenous injection, then, the results showed that there was a significant difference in body weight. However, from the results of H&E staining, tumors can be found from the lungs of both mice. In conclusion, the results of the study showed that Helios knockout in gastric tumor cell increased the ability of migration and invasion in vitro, but in the subcutaneous injection in nude mice, the result showed that tumor appearance is relatively slow in Helios knockout gastric tumor cell; however, with or without Helios knockout tumor cells showed tumor appeared in lung after in intravenous injection. However, in the previous studies, it has been pointed out patients with gastric cancer who have low expression in Helios have poor prognosis, especially in patients with Helicobacter pylori infection and advanced gastric cancer. However, in the experiment of my study, it has not treated the induction of Helicobacter pylori infection in much detail, thus, the ability for cell metastasis in animal experiments needs to be confirmed by more research or further experiments.
