Summary: | 碩士 === 元培醫事科技大學 === 生物科技暨製藥技術系碩士班 === 106 === This study investigated the effect of plant growth regulators on Lilium micropropagation with various different concentrations of Auxins and Cytokinins on the regeneration and proliferation of regenerated bulblets. Two different lilies, Lilium Hybrid ‘Casa blanca’ (Designated as: Casa blanca), and Lilium Hybrid ‘Pink Mist®’ (Designated as: Pink Mist) were studied. The results of experiment involved induction of callus, bulblets regeneration, proliferation, induction of roots formation, and acclimatization of micropropagated plants were observed. Totally 117 combinations of 2 Auxins (NAA, 1-Naphthaleneacetic acid; IAA, Indole-3-acetic acid) and 3 Cytokinins (TDZ, Thidiazuron; BA, 6-Benzylaminopurine; KIN, Kinetin) with various concentration (0, 0.1, 0.2, 0.5, 1.0 mg/L) were applied to culture media. The basal medium consisted of half strength MS medium supplemented with 10 mg/L thiamine-HCl, 100 mg/L myo-inositol, 1 mg/L pyridoxine-HCl, 1 mg/L Nicotinic acid, 30 g/L Sucrose, 1 g/L Activated charcoal, and 4 g/L Gelrite. The plant materials were all from the stem of mother bulbs that were cut into smaller segments and used as the explant for micropropagation. The culture condition was room temperature in the darkroom. The best result from single plant growth regulator treatments for root induction of Casa blanca and Pink Mist explants were NAA 1.0 mg/L and 0.5 mg/L, respectively. For the best Cytokinin treatments in bulb regeneration and roots formation were TDZ 0.5 mg/L and 1.0 mg/L for Casa blanca and Pink Mist, respectively. For the results from the combination of plant growth regulator treatments for 2 lilies: (1). Casa blanca treated with growth regulator combinations (NAA/ Cytokinins): The best result regeneration bulblets was NAA 0.1 mg/L and TDZ 0.2 mg/L, and the optimized rooting treatment was NAA 0.2 mg/L and KIN 0.2 mg/L. Another Auxin combined with Cytokinins (IAA/ Cytokinins), for bulblet regeneration was IAA 0.1 mg/L and KIN 1.0 mg/L. For roots formation was IAA 1.0 mg/L and BA 1.0 mg/L. (2). Pink Mist treated with growth regulator combinations (NAA/ Cytokinins): For regeneration bulblets and roots formaiton was NAA 1.0 mg/L and TDZ 0.1 mg/L. Another Auxin combined with Cytokinins (IAA/ Cytokinins), for bulblets regeneration was IAA 0.5 mg/L and KIN 0.5 mg/L. For roots formaiton was IAA 1.0 mg/L and TDZ 0.1 mg/L. Regenerated plantlets were subjected to acclimation after 20 weeks in vitro culture. The survival rate was up to 80% after 6 weeks and all plants were morphologically normal. This study provides informations of hormones alone or combinations for regeneration bulblets in Lilium micropropagation and acclimatization of regenerated plants.
