Abstract
In the present study, we used sesamin derivatives, 3-bis (3-methoxybenzyl)-butane-1,4-diol, (BBD) as the subject, and analysis its protect effect from injury from irradiation on retinal pigment epithelial cell (ARPE-19 Cells).The cell viability (MTT) and cytotoxicity (LDH) were measured after incubation with the BBD for 24 hours and then subjected to irradiation with ultraviolet light (100 MJ). Apoptosis of ARPE-19 Cells was induced by UV irradiation (100 MJ), and reactive oxygen species (ROS) were generated. The protein expression of MAPKs, Caspase-3, and Bax was analyzed by Western blotting. The results showed that the cell viability (MTT) of ARPE-19 Cells was significantly decreased after UV-stimulation, and the cell viability was significantly increased with dose-dependent of BBD concentration. Our study showed that BBD pre-treatment can protect RPE cells from UV-induced apoptosis. The present study suggests that the BBD in the prevention of retinal diseases associated with UV-stimulation induced oxidative stress such as AMD disease.
