Summary: | 碩士 === 國立雲林科技大學 === 化學工程與材料工程系 === 106 === This research uses Poly (HEMA‐PEGDA-MAA) hydrogel by UV-crosslinking to entrap Lactobacillus brevis RK03 and uses MSG as cultivation medium material to make MRS cultivation medium for the fermentation and production of γ-Aminobutyric acid (GABA).Through the control of hydrogel proportion and balance of mechanical properties with water content the research finds the most suitable hydrogel for the growth of L. brevis RK03. Experimental results show that 1500P5M5 hydrogel with HEMA as main monomer, 5 % Methacrylic acid (MAA), 5 % Polyethylene glycol diacrylate (PEGDA)-1500, 1 % Benzoin isopropyl ether has an excellent tensile strength of 1.23±0.01 MPa, Elongation at break 219.8±10.8 % and Water content of 44.53±1.33 %.
Using this hydrogel in immobilized batch fermentation, we obtain a fine GABA yield of 445.0±14.2 mM, a conversion rate of 98.89±3.16 %, and a MSG residue of merely 0.409±0.02 %.Proceeding on with continuous fermentation, the hydrogel had a conversion rate of 95 % after 5 times usage of GABA, 90 % after 10 times, and 86.15±2.50 % with the 11th experiment. Therefore 5 % PEGDA-1500,5 % MAA hydrogel has great potential for development in the use of industrial production.
