In Taiwan, the mortality rate of oral cavity cancer is the fourth of all cancers. Warburg effect indicates that cancer cells gain ATP by rapid anaerobic glycolysis to maintain fast proliferation. When cancer cells start the metastasis, cells occur EMT. This study investigates the metabolic change of Twist1-induced EMT cancer cells. First, we used SAS cell lines of HNSCC, and knockdown Twist1 at the cell line that makes SAS cells transit to epithelial type. We detect the change between the wild type and shTwist cell line in metabolism with XFe24 Extracellular Flux Analyzer. From the result, OCR and ECAR are both lower in shTwist1 cell line than wild type. Then we compare the metabolites level of shTwist1 cell lines with wild type. The acetyl-CoA level is not different in shTwist1 cell line, but the lactate level is increase in SAS-shTwist1 cell line. Otherwise, succinate is accumulated in Twist1 Knockdown SAS cells. Finally, we analyze some genes and proteins about TCA cycle and biosynthesis of amino acid and fatty acid. The result shows PDK1 is increase and it will translocate into mitochondria to inactive PDH, and then effect TCA cycle in Twist1 knockdown SAS cell line. Muh-Hwa Yang S.J.
