Methods: Total 56 OVX rats were divided into 5 groups: sham operation (sham operation, SHAM, N=10), ovariectomy (ovariectomized, OVX, N=11), resistance training (OVX+RT, N=12), night time eating high fat (OVX+NF, N=11), and night time eating high fat and resistance training (OVX+NF+RT, N=12). The five groups were given a normal diet everyday. The resistance exercise group were trained for 8 weeks, each training interval of 48 hours. The high fat night time eating group were given high-fat diet after exercise resistance. At the seventh week of exercise intervention, the oral glucose tolerance test (OGTT) was performed. The last exercise of muscle strength test was performed after exercise intervention. 72 hours after resistance intervention, we would take the rats’ lexor digitorum profundus (FDP) and flexor hallucis longus (FHL), abdominal fat, and blood. Taking the rats’ FHL to analyze the expression of LC3, p62, Beclin 1, AMPK, pAMPK, STAT1, pSTAT1, STAT3, pSTAT3 and FOXO3a were measured. The levels of IL-6, TNF-α, IFN-γ and Estradiol in blood and CD68, CD163 in FHL were measured. And taking the FHL to analyze and calculate the muscle cells, cell nucleus, and cross-sectional area. Results: Compared to night time high fat diet, ovariectomy rats that was done resistance training plus high fat diet at night were muscle mass, muscle strength, body weight, fat mass, blood glucose, and insulin were improved, but total daily calorie intake was no significantly different. The concentration of IL-6 in the blood increased the expression of STAT3 protein to produce muscle hypertrophy, the concentration of TNF-α in the blood was decreased, the activity of CD68+ and CD163+ cells in muscle were not different, and the expression of autophagy-related protein were increased. Conclusion: Menopausal rats with resistance training accompanied by a nighttime high-fat diet can improve muscle mass and muscle strength and reduce fat accumulation.
