If stem cell differentiation and in vitro culture used as a source of skin cells, the remaining autologous skin can be preserved. In this study, we take 1. SHEDs induced differentiation into epidermal stem cells. 2. Combine epidermal stem cells with two materials to perform a rat wound healing test.3. TISCHEL Solution for sealant, a mixture of epidermal stem cells mixed was used for wound healing test in rats. Based on the experimental results, we found that the induction of the induction medium to the third day of SHEDs differentiation into epidermal stem cells. And choose two materials on the dressing: 1. Scaffolds group: the used of PVA in combination with Fibroin on the dressing. Fibroin promoted collagen regeneration and accelerates wound healing. 2. Gel group: Hydrogel in combination with the extracts of Ampelopsis, used the anti-inflammatory properties to accelerate wound healing. In the animal experiment, the wound healing was observed. Comparison of simple materials in the Gel group on the third day to unobserved inflammatory redness reaction, and began to wound contraction. The fifth day, the best was wound closure of 41.71%. And the rate of wound healing is about 5% higher than TISCHEL solution. Comparison of combine stem cells with materials groups. Granulation tissue regeneration was performed on third day in the Scaffolds+ESCs group. By the fifth day, the best was wound closure of 53.49%. And the rate of wound healing is about 5% higher than the combine stem cells with TISCHEL Solution. The above data shows that Fibroin and the extracts of Ampelopsis has the effect of accelerated wound healing.
