It is a perennial erect herb, a Berberidaceae Epimedium, which is commonly used in traditional Chinese medicine for warming kidney and strengthening the yang, strengthening the bones and removing rheumatism. This experiment will focus on the part of the strengthening the bones. After breaking the Epimedium, we passed the 200mesh sieve and the powder was stored in a dry and ventilated place. We extracted the Epimedium powder in three ways, traditional extraction, static extraction and supercritical fluid extraction. The extracted extract was dried to powder by a lyophilizer and stored at -4 °C. The extract powder was dissolved in α-MEM by DMSO, diluted with a gradient concentration, and the optimum concentration of each extract powders was measured by the MTT method, then begin long-term culture. In the culture, alkaline phosphatase, calcification, immunofluorescence staining, qRT-PCR and other methods were used to test the bone differentiation performance at 7, 14, 21, and 28 days, respectively. The results showed that the water extract had poor osteoinductive effect, and both the alcohol extract and the supercritical flow extract had good induction effects, while in the calcium deposition, the supercritical extract had the best induction effect.
