It has been identified as being overexpressed in human lung, breast, prostate, and ovarian cancers. Previous several studies demonstrated that TPD52 gene promotes lipid storage in lipid droplets by increasing lipid droplet proteins on lipid droplets. Besides, overexpression of the TPD52 gene in 3T3-L1 cells in mice results in significant increases in triglyceride, free fatty acids, and adipogenesis, but can't determine its mechanism, whether it will increase adipogenesis by affecting the volume or number of adipocytes? If it increases the number of adipocytes, does it affect PPARγ, C/EBPα, and C/EBPβ and other adipocyte differentiation related genes. Therefore, this study hopes to establish a large number of zebrafish model organisms that express the TPD52 gene. Through the promoter of β-actin, it expresses the whole body and collaborates with the tet-off system to find out the ways in which TPD52 gene alters lipogenesis and its mechanism. During the experiment, we used Oil Red staining and dissected on TPD52 transgenic fish to determine that TPD52 gene actually increased fat production. In the tissue paraffin sections we found that the number of TPD52 adult fish adipocyte was significantly more than Wild-type adult fish, determine that TPD52 gene increases adipocyte differentiation. Finally, we extracted Wild-type adult fish and TPD52 transgenic fish adipocyte Total RNA and then conduct semi-quantitative and rt-PCR, we observe that adipocyte differentiation related genes (For example: PPARγ, TGFβ, STAT5A, and GATA3) does have a significant difference from wild-type adult fish, Therefore, we can determine that TPD52 gene promotes the differentiation of adipocytes by regulating adipocyte differentiation related genes, it proves that TPD52 gene plays an important role in adipogenesis.
