In the chip preparation part, photo resist SU-8 GM1075 is used to prepare the micro channel chip mold using photo-lithography process technology. Moreover, casting technique is used for molding, connected to glass substrate with platinum, finlly, biological chip of this thesis is then prepared. In the system architecture preparation, green laser of wavelength 532nm is used, and high pass filter of wavelength 550nm is also used. Meanwhile, AutoCAD and PSoC are also used to prepare experiment system circuit and to make experimental platform for test system. In the microbial culturing aspect, edwardsiella and coli bacillus of different fragment are adopted to perform biological hybridization comparison. In the test aspect, this experiment contains fiuorescence test, which is used to analyze the electrical signal of microbe hybridization, though this bio-chip of test analysis experiment, and though the specificity on microbe from fluorescence probe HEX, then electrical signal can be used to distinguish whether the microbial hybridization is a success or not. Meanwhile, this experiment not only reduces the test time, it also greatly reduces the cost of the test system, moreover, it is very easy to carry, therefore, in the future, it is expected to have wide application in bio-medical test area.
