It is known commercially as Huangqin is often used for boosting immunity, this plant-rich of flavonoid compound with antioxidant properties. The aim of this study was to examine the survival rate, innate immune responses, immune genes expressions and resistance against Lactococcus garvieae. In the present study, M. rosenbergii injected with aqueous S. radix water extract at 1, 5 and 10 µg/µl. Total hemocyte count (THC), phenoloxidase (PO) activity, respiratory bursts (RBs) activity, transglutaminase (TG) activity and lysozyme activity of hemocytes were conducted to assess the immunological responses after reverse-gavage feeding. The results showed that after 3 and 6 hours of reverse gavage feeding with aqueous S. radix extract at 5 µg/µl had increased significantly of immune response compared with the control group. THC of the prawns was increased significantly post RGF, and the highest value of THC was observed at 5 µg/µl of S. radix. For immune responses, PO, RBs, and TG of prawns injected with 5 µg/µl of S. radix water extract using RGF were significantly increased and only lysozyme activity significantly increased by 10 µg/µl, however, all immune response activities of the prawns return to normal 12 hours post RGF except lysozyme that is still increasing until 24 hours. For immune-related genes, LGBP, proPO, and peroxinectin increased 12 hours post-reverse gavage feeding and back to normal on 24 hours. Related to survival rate, the highest percentage of survival rates in prawn was observed at 5 µg/µl of S. radix water extract, compared with other group and control group challenged with L. garvieae. These study concluded and recommended that S. radix water extract at 5 µg/µl can be used as an immunostimulant and additional substances to enhance immune responses and immune genes expressions in M. rosenbergii using RGF.
