Description
Summary:碩士 === 國立屏東科技大學 === 生物科技系所 === 106 === Protein-protein interaction, especially protease and its inhibitor, have been studied using the three-dimensional structure of protein complexes and binding affinity data to obtained their functional information. Based on previous study, sesame asparaginyl endopeptidase (SiAP) and cysteine protease inhibitor (SiCYS) have been successfully isolated and transformed to competent cell. The predicted SiCYS structure displayed a polar contact with one of the papain active site. Meanwhile, the predicted SiAP structure showed a steric effect that blocked the SiCYS to inhibit its active site. Expression analysis of SiAP indicated that this gene was not expressed during germination. In addition, the results of functional assays indicated that SiCYS might lead to the apoptosis of RAW 264.7 cell.
