Summary: | 碩士 === 國立嘉義大學 === 森林暨自然資源學系研究所 === 106 === Chinese Junipers (Juniperus chinensis cv. Shimpaku) are in the genus Juniperus of the family Cupressaceae. Because of lack of pollination, low pollen viability and embryo degeneration, the seed germination of Junipers is extremely low. The air layering is used predominantly to propagate while this experiment was conducted using warm and cold stratification on seed germination. In addition, this study explores the influence of dark septate endophytic fungi (DSE) and indole butyric acid (IBA) on the rooting effectiveness of air layering propagation for Chinese Junipers, using different concentrations of IBA and different species of the DSE. Lastly, this research discusses if the DSE vary physiologically and biochemically. After the treatment of warm and cold stratification of Chinese Junipers seeds, the germination percentage was only 18.2%. In experiment I of air layering propagation, the IBA (2,000 ppm IBA) was found most effective for rooting and the CkDB5 strain treatment came second. In experiment II of air layering propagation, it was discovered that, with different concentrations of IBA (0, 1000, 2000 and 3000 ppm), the number of roots in both 2,000 ppm IBA and 3,000 ppm IBA was significantly higher than the control group (p<0.05). When treating with different DSE species (MoAL5, CkDB5 and RrtHH10 strains), all of the inoculations with DSE reached up to the rooting percentage of IBA treatments (2,000 ppm IBA). Yet, of the 8 experiments with different IBA concentrations and DSE species, both the group treated with CkDB5 strain and that with 2,000 ppm IBA + CkDB5 strain received significantly different than the other treatments in terms of the average number and length of roots. According to the analysis of the physiological and biochemical characteristics of DSE, the three strains, are capable of producing IAA. Second, the analysis of protease indicates the RrtHH10 strain is capable of producing protease and is using organic protein. Third, in terms of the ability to dissolve organic phosphorus, none of the three strains can. The result of this study, the germination of Chinese Junipers seeds is not easy. Hence, when conducting air layering propagation, it is suggested to us the CkDB5 strain instead of IBA rooting concentrate to boost rooting because this strain can not only stimulate root development but help the host plant grow.
