In the past, dextrose injection is often used to relieve pain in torn joints and ligaments. This study aims to investigate the effects of both dextrose and dextrose hydrogel injections on muscle Contusion and whether dextrose hydrogel injection is better than dextrose injection. Methods: 80 male ICR mice were randomly divided into 10 groups, 5 groups were uninjured control and 5 groups were simulated with muscle injury by mass drop injury (MDI) and all the groups were supplemented with either 5% or 10% dextrose and dextrose hydrogel. The experimental animals sacrificed one week later of the intervention. blood test and H&E-stained section will be observed to evaluate the inflammatory condition. Results: the results revealed that no significant difference in body weight, feeding status and drinking status were observed during the experiment (P>0.05) . Although the metabolic biochemical values showed signs of differences (P<0.05), however, these can be deemed as individual differences. The H&E stain further revealed that groups with 5% and 10% dextrose hydrogel injections showed signs of tissue necrosis and macrophage aggregation, no matter simulated with muscle injury or not. Conclusion: In summary, both dextrose and dextrose hydrogel injections will not affect the metabolism of liver/kidney, hormone and other metabolic biochemical values. However, through pathological observations, dextrose hydrogel injection will induce tissue necrosis and macrophage aggregation instead. Therefore, dextrose prolotherapy is preferred as the substances is safe and can be absorbed by the body. As the results of the experiment did not show significant difference between the 5% or 10% dextrose injection, it is suggested that different variants can be taken into account in future research, including inspection time, the structure of hydrogel etc. In hope of finding better healing methods.
