It can be activated by ligand binding and then translocate into nucleus to activate specific target genes. PPAR γ is ubiquitously expressed in adipose tissue, and plays important roles in adipogenesis, lipid metabolism, and glucose homeostasis. Previous studies show that macrophages and dendritic cells (DCs) also express PPAR γ; however, the role of it on DC function awaits further investigation. Aim: To study the effect of PPAR γ activation on DC function. Method: Murine bone marrow derived dendritic cells(BM-DCs) were treated with ciglitazone, a PPAR γ agonist for 8days, and then stimulated with LPS for 24 hours. The cells were analyzed their maturity and cytokine pattern using flow cytometry and ELISA, respectively. The lung inflammation model was established for analyzing DC function. Results: The results showed that long-term treatment to PPAR γ agonist significantly inhibited DC maturation and altered cytokine pattern. Conclusion: PPAR γ activation may alter DC function, and then affect immune response.
