To diagnose and treat colorectal cancer earlier is the cornerstone of preventive medicine. Besides, it’s also popular to search the specific biomarker that can predict the sensitivity colorectal cancer to the current chemotherapy regimen, further correlating the clinical outcomes. According to the current treatment guideline, Oxaliplatin and Fluorouracil is the choice of first-line chemotherapy regimen, but there is still no reliable biomarker can help us to predict the treatment efficacy. The biomarker Bmi1 had been widely known to be involved in the tissue proliferation and differentiation, recent works shown that it also played an important role in tumorigenesis via cell cycle regulation, cancer stem cell activation and DNA damage response. Our study aimed at discussing the relationship between of Bmi1 and colorectal cancer. We used immuno-histochemical staining of clinical tissue specimen, validating that the expression of Bmi1 was positively correlated with colorectal cancer stage and survival outcome; also used cell line HCT116, exploring the expression of Bmi1 influenced the response to genotoxic agents; Finally, we identified downstream stem cell-associated transcription factor including Klf4, Oct4, Sox2 and Nanog were regulated by Bmi1 by microarray.
