Excessive absorption of UV will generate free radicals that cause injury or inflammation of the skin and scalp, and lead to aging or even cancer. The main purpose of this study was to investigate the antioxidative effects of extract from Chlorella pyrenoidosa and to investigate the effects of the extract on the growth of mouse fibroblast (3T3) and human hair follicle dermal papilla cells (HFDPC) and the mechanism of protection against ultraviolet radiation. The results showed that the extract of C. pyrenoidosa had good antioxidant activity including ability to scavenge superoxide radicals (EC50 0.22%), reducing power (EC50 4.03%) and ability to chelate ferrous ions (EC50 2.11%). The mechanisms of C. pyrenoidosa extract that can promote the growth of 3T3 and HFDPC cells may be relate to the activation of Akt and ERK. C. pyrenoidosa extract also has a significant protective effect on the cytotoxicity induced by different UVB energies, but it inhibits the activation of ERK. It also has a good effect on the cell wound healing test, and its effect may be related to the induction of matrix metalloproteinases (MMPs) activity, especially in UV-irradiated cells, MMP-2 activity was significantly increased. In summary, the extract of C. pyrenoidosa with antioxidant capacity, promoting cell growth and reduce cell damages of 3T3 and HFDPC.
