Summary: | 碩士 === 國立中正大學 === 機械工程系研究所 === 106 === This study developed a grating-coupled biosensor and combined the sandwich ELISA to detect Pseudomonas aeruginosa drug channel protein (OprD).Because the hospital's current identification method for the resistance of Pseudomonas aeruginosa takes a long time, if it is measured by other biosensors at a low concentration, it must have high sensitivity performance. The sensing method of this study has the characteristics of high sensitivity and specificity. In addition to increasing the specificity of the immune response and amplifying the response signal by the sandwich ELISA. The evanescent wave field depth of the grating sensor and the small molecule test object size matching problem can be simultaneously improved and effectively improve the detection limit of the sensor. In the study, the types and concentrations of surface modification molecules on chips were also discussed. It is expected that stable and high-efficiency sensing chips can be obtained by better modification conditions. In addition, since the grating-coupled biosensor of this study utilizes the effective refractive index of the surface of the detection grating as a detection mechanism, it belongs to an open biosensing platform. By modifying different antibody molecules, a variety of analytes can be detected, and the sandwich ELISA can improve the sensing performance, so it has the potential to develop into a clinical detection system.
