The purpose of this study is to discuss the sampling timing of non-point source pollution and establish the localized pollution load coefficient by using the phenomenon of first flush. In this study, non-point source pollution of different land use rainfall events was collected, and a total of 11 rainfall events, traffic landuse (Jianguo Viaduct), agricultural landuse (dragon fruit farmland, bamboo shoots farmland) and watershed of Nan-Zi-Gou river, respectively. The effects of suspended solids, chemical oxygen demand, ammonia nitrogen and total phosphorus on the first flush were studied, then the M(V)curve was used to analyze whether the first flush of the quality was related to the characteristics of rainfall, the land use pattern and environmental factors. The sampling time of non-point source pollution is discussed, and the unit pollution load coefficient of different land use is deduced by unit rainfall load method as the basis of domestic localization data. The results show that, whether in the high impervious area of the traffic landuse, or in the low impervious area of agricultural landuse, have the phenomenon of mass firat flush, which traffic land (Jianguo Viaduct) in the rainfall reached 6±3 mm, in addition to ammonia, the other pollutants have moderate to mild of the mass first flush phenomenon, agricultural landuse (dragon fruit farmland, bamboo shoots farmland) in the rainfall reached 17±5 mm, the suspended solids have a high of the mass first flush, chemical oxygen demand, ammonia and total phosphorus there is moderate to mild of the mass first flush phenomenon, watershed of Nan-Zi-Gou river due to the larger catchment area, the time of concentration is longer, only in the middle of the rain event, part of the pollutants began to appear the phenomenon of first flush, so there is no phenomenon of mass first flush. In planning non-point source pollution sampling plan, suggestions can refer to the timing of the mass first flush.
