Description
Summary:碩士 === 亞洲大學 === 生物科技學系 === 105 === AC-10 is a well-known traditional Chinese medicine in Taiwan. AC-10 possesses many bioactivities including anticancer activity. AC-10 showed anti-breast cancer effect induced breast cancer cell apoptosis, however, the metastasis was not clearly studied. TGF-β is a pleiotropic, anti-inflammatory cytokine and a pivotal regulator of a wide range of cellular processes, including cell cycle, proliferation, differentiation, epithelial-to-mesenchymal transition (EMT) and metastasis. In cancer cells, EMT confers tumor cells with a distinct advantage for metastatic dissemination. In this study, MDA-MB-231 cells were treated with AC-10 (0-60 μg/mL) for 24 h. The protein levels of E-cadherin in whole cells were determined by Western blotting. AC-10 up-regulates E-cadherin expression. Results showed that AC-10 inhibited EMT via modulation of Wnt/β-catenin and inhibition of Smad2/3 signaling pathways on triple-negative breast cancer (MDA-MB-231) cells in vitro and in vivo.
