H. Shyu, Ph.D. The Contents of Abstract in This Thesis : Sesamum indicum L. is an ancient and important oil seed crop grown widely in tropical and subtropical areas. Sesame seeds storage large amounts of triacylglycerols (TAGs), which are hydrolyzed to free fatty acids and glycerols by TAG lipases during germination. The glycerol is phosphorylated and then enters gluconeogenesis pathway for the generation of glucose. The free fatty acids are activated to acyl-CoAs and then enter the -oxidation pathway, which produces the energy for sesame seedling growth following germination. For the study of genes related to lipid metabolism, three hydrolase-like genes, named SiLip1, SiBioH, SiMES20, are cloned from sesame during germination. A putative monoacylglycerol lipase cDNA, SiLip1, which is similar to Saccharomyces cerevisiae ROG1, contains an open reading frame of 1,080 bp encoding a protein of 360 amino acids. SiBioH cDNA contains an open reading frame of 1,023 bp encoding a putative pimeloyl-ACP methyl ester carboxylesterase of 341 amino acids. SiMES20 cDNA contains an open reading frame of 606 bp encoding a putative inactive methylesterase 20 protein of 202 amino acids. Study on the function of three hydrolase-like proteins expressed during seed germination will provide clues to understand their physiological roles in lipid metabolism.
