Chewing betel nut, smoking, and alcoholism are the three major risk factors which may contribute to the development of HNSCC. Recent reports have demonstrated that long-term exposure to either nicotine derivatives or arecoline may be associated with tumor progression of HNSCC.   In this study, the cell model of long-term exposure to nicotine derivatives and arecoline was established. A comparative analysis using SCC-25 and Fadu cells with or without nicotine derivatives and arecoline exposure was performed to evaluate cell proliferation, migration, and invasion as well as the expression of epithelial-mesenchymal transition (EMT) marker, drug-resistance gene expression, cancer stem cell properties, and anti-apoptotic activity.   As a result, either long-term nicotine derivatives or arecoline exposure, or exposures to both risk factors was found to stimulate cell proliferation, enhance migration and invasion, induce EMT with morphological alterations in a dose-dependent manner, increase stemness, and attenuate apoptosis.
