In order to clarify the roles of nitric oxide (NO) whether involved hypertension induction and progressing. The N(G)-Nitro-L-arginine-methyl ester (L-NAME, a NO synthase inhibitor) have be used in this study. The C57BL/6JNarl male mice were administrated L-NAME 80 mg/kg (LN group) or drinking water (C group) by oral gavage for 4 weeks. The weight changes, metabolic rates, blood pressure and histopathologic changes were evaluated. Our results showed that significant difference in energy expenditure, O2 consumption, CO2 produced, respiration quotient and activity between LN group and C group. In LN group, the blood pressure increased 22 mmHg (P< 0.05) than in water group. Moreover, the angiotensin II concentration increased 404.9 mg/dl (P< 0.05). The significant reduction of serum albumin level in LN group was observed. The inflammation response and slightly fibrosis were observed in the heart and kidney of LN group. In conclusion, the present study have successfully established L-NAME induced hypertension model on the mice. It will provide a good animal platform for novel hypertension drugs verification.
