After that PDCoV has been reported in piglet diarrhea in United States (2014), South Korea (2014) and Thailand (2015). To determine whether this virus also existed in Taiwan, a molecular surveillance was conducted using IDEXX real-time PCR kits. There are 112 rectal swabs from diarrheal piglets were collected from the middle and south Taiwan in last year. Another two coronaviruses causing piglet diarrhea, that is porcine epidemic diarrhea virus (PEDV) and transmissible gastroenteritis virus (TGEV), are also examined in this study. The real-time RT-PCR results showed that detection rates of PDCoV, PEDV, and TGEV are 23.2% (26/112), 21.4% (24/112), and 1.8% (2/112), respectively. One of the 112 specimens (0.9%) is all three coronaviruses detected and 12 of them (10.7%) might be PDCoV and PEDV co-infected. After cloning and sequencing, three nucleocapsid (N) genes of PDCoV were analized by MEGA7. The phylogeny results showed that the N genes cloned in Taiwan are most similar to those isolates from United States. The bacteria expressed and purified nucleocapsid protein of PDCoV was also used as antigen coated on microtiter plates for serological survey. Swine sera tested were collected from 2011 to 2015. The ELISA results showed that 279 of 1,039 (26.9%) sera can react with the PDCoV nucleocapsid protein. All this implied that PDCoV may be existed in Taiwan for a while.
