In this study, triterpenoids metabolites of Tuber borchii were produced in the submerged culture. In order to enhance triterpenoids production, various additions such as palmitic acid, stearic acid, linoleic acid, chitosan, CaCl2 and limonene were tested. It was found that adding 1 g/L linoleic acid to the medium could elevate the triterpenoids production to 129.29±20.5 mg/L, which was 2.94-fold to the control. To find the optimal conditions for growing mycelia in solid state and submerged cultures, factors such as carbon and nitrogen sources and concentrations, potassium ion, and magnesium ion levels were considered. The results showed that the optimal medium for the solid plate was as follows:glucose, yeast extract, malt extract were 13, 8.68 and 8.51 g/L, respectively. The highest mycelial diameter obtained under the optimal conditions was 6.87±0.26 cm, which was 3.97-fold increase to the control. In the submerged fermentation, according to the response surface methodology, the optimal concentrations for glucose, yeast extract, peptone, malt extract, KH2PO4 and MgSO4.7H2O were 19.45, 4.58, 7.91, 5.3, 0.58 and 0.82 g/L, respectively. The validation tests showed the experimental values were in good agreement with the predicted values. The highest dry cell weight (2.98±0.18 cm) was obtained under the optimal conditions, which was 1.39-fold higher than the control. At last, the addition of 1.5 g/L linoleic acid at day 14 to the optimal medium could elevate the triterpenoids production to 212.63±16.58 mg/L, which was 4.84-fold increase to the control.
