Summary: | 碩士 === 義守大學 === 化學工程學系暨生物技術與化學工程研究所 === 105 === Because of the development of many diseases are related to the immune abnormalities, and there is no relevant literature that explored the immune system of banana peels in the past, so this research is to explore the remission of acute inflammation and the separation of active principle for Musa paradisiacal peel n-butanol extract(BPE).In this study, we divided the BALB/c mice into four groups; the control group, the low-dose and the high-dose BPE group (42; 166mg / kg BW) and the positive control group-PDTC group. After a week of feeding water and sample for each group, we do the intraperitoneal injection of LPS (Lipopolysaccharide) to induced acute inflammation, the mice will sacrifice 3 hours later, and we collected the blood, organs and lymph nodes for immuno-biochemical analysis.The results shows that compare to the control group, the levels of blood IL-6 and TNF-α in the high-dose BPE group were significantly decreasing, which is the same as the control group. The amount of the IL-6 peritoneal cell secretion was also significantly decreasing, and when observing the stimulation of the spleen to T cell, we found that the secretion of IFN-γ and IL-6 were also decreasing. But instead, the formation of IL-4 and IL-10 has increased slightly. The research shows that BPE adjust the balance of Th1/Th2 and reduced the response of inflammatory. And in the adjustments of T cells (Regulatory T cell), the high-dose, low-dose group and the positive control group were significantly decreased and were having significant differences.
In this research, we analyzed the anti-inflammatory effects of the components in BPE and found that, 5-hydroxymethylfurfural, Guaiol and the oleamide concentrations of 100μM can inhibited the ability of LPS stimulating macrophage to secret IL-6 And TNF-α.
This research shows that BPE and its isolated components can inhibit LPS to induce acute inflammation by decreasing the formation of the inflammatory cytokine.
