In this study, Salmonella, Clostridium difficile, Staphylococcus aureus, and Campylobacter were dectected in environmental water in Taiwan. The PCR products were used to detect from this strains and the wastewater of livestock poultry from Puzi River which were collected. The influence of the monitoring point on Puzi River was studied. The results showed that the detection rate of Salmonella is 6.14% in domestic and livestock wastewater, 3.13% in Puzi River, and 0% in coastal areas and aquaculture water. The rate of the detection of Clostridium difficile in domestic and livestock wastewater is 9.48%, in Puzi River is 1.04%, in coastal areas and aquaculture water is 0%. The rate of the detection of MRSA in domestic and livestock wastewater is 7.89%, in Puzi River is 0%, in coastal areas and aquaculture water is 0%. Campylobacter to the detection rate is all 0%. This study focused on which are the use of PFGE as a standard method for Salmonella compared ERIC-PCR and MLST that the accuracy of two classification methods. The results of ERIC-PCR typing of Salmonella environmental sample showed that most of serotypes were classified in dendrogram, and we found the detection was fast and time-saving, and similarity between strains can be done with the analysis of known strains and doing simple typing. The advantage of using MLST typing is that the data can be exchangeable and used to identify more accurate serotypes, but the method is time-consuming,laborious and expensive. In this study, the ERIC-PCR dendrogram of C. difficile was also typed and compared with its toxic gene. The results show that more information is required to enhance the typing effect.
