Preconcentration is an important step that increases accuracy of subsequent detection, especially for samples with extremely low concentrations. Because of overlapping electrical double layers in the nanofluidic channel, the concentration polarization effect can be generated by applying an electric field. In this study, the preconcentration chip for proteins was fabricated using simple standard soft lithography with a polydimethylsiloxane (PDMS) replica. Using various concentration gold nanoparticles (1.325、2.65 and 26.5 nM) deposited on glass substrate by self-assembly. The results showed apparently that all case using 15 nm gold nanoparticles for SAM achieved protein concentration successfully. The experimental results indicate that gold nanoparticles concentration do not affect concentration results. Over concentration gold nanoparticles (26.5 nM) have regular pattern and compact structure. A protein sample with an concentration of 10 μM was concentrated to 50-fold in 30 min by using the proposed chip.
