Through the catalytic action of intracellular 6G-fructofuranosidase, sucrose is converted into neokestose, neonystose and a small amount of 1-kestose. Neofructooligosaccharides (neo-FOS) is an important prebiotic, which can promote the proliferation of intestinal bifidobacteria, and thus enhance human health. In this work, fermentative production of 6G- fructofuranosidase was conducted using a 5-L fermenter with a working volume of 3 L under various conditions including pH, agitation, aeration and concentrations of carbon and nitrogen sources. The optimal conditions for 6G-fructofuranosidase production were: 21℃, pH 7, 700 rpm and 2 vvm. The fermentation medium consisted of the following ingredients per liter: yeast extract 30 g, ammonium nitrate 3 g, dipotassium phosphate 5 g, and magnesium sulfate heptahydrate 0.5 g, and every 50 g sucrose per liter was added at 0 and 24 h of fermentation. The fermentation was complete at 44 h and the highest 6G-fructofuranosidase activity at 284 U/mL was obtained.
