Summary: | 博士 === 國立臺灣科技大學 === 應用科技研究所 === 104 === Poly lactide-co-glycolide acid (PLGA), approved bio-degradable polymers by the United States Food and Drug Administration (FDA), can be used to make injections for human. It has a widely applied in control release of the active substance, targeting, prolong the half-life of drugs, increase bioavailability in vivo and other characteristics. PLGA microspheres is used in studies of drug delivery systems and immunology, which are benefit for increasing the stability, antigenicity, and good immunoadjuvant. In this study, varied microspheres encapsulated technology was developed to preparing different microspheres and to get the experimental parameters, which included with different ratios of PLGA and PEG as the core materials, and use BSA as a model antigen. Then microencapsulated of dengue NS1 or Yersinia pestis F1 antigen microparticles were successful prepared. The animal study showed that a high antibody titers by single injection, and provide complete protection to immunized animals. The resulted provied that a high-efficiency induced antibody technology platform to facilitate the preparation of the specific antibody was developed, and the antigen encapsulated micrparticles have potential further to develop a new vaccine.
