Many studies indicated that DEHP was associated with developmental, reproductive and hepatic toxicity in animals and humans. Curcumin (CUR) is a phytochemical derived from rhizome of curcuma longa, present in the curry spice. CUR has exhibited anti-inflammatory, anti-infective and antioxidant properties. Methods: Experiment 1: Male 42-postnatal day (PND)-old Sprague Dawley (SD) rats were randomly divided into control, 1 mg/kg, 2 mg/kg, and 8 mg/kg groups to establish the dose-response assessment of CUR. Experiment 2: Animals were divided into control, CUR group (2 mg/kg), DEHP (1000 mg/kg), and CUR+DEHP (CUR 2 mg/kg + DEHP 1000 mg/kg) groups to evaluate the protective effects of CUR on DEHP-induced sperm dysfunctions. The change of body weight and amount of drinking water were recorded from PND 42 to PND 105; at the end of the experiment, all rats were euthanized and their testes and sperm functions were analyzed. Results: In experiment 1, the sperm super oxide anion levels of rats in the CUR 2 mg/kg group significantly decreased. In experiment 2, we found relative testis and epididymis weights in the DEHP group significantly decreased compared with those of control, CUR, and CUR + DEHP groups. The sperm super oxide anion levels of in the CUR and CUR+DEHP groups significantly decreased compared with those of their controls, respectively. Sperm chromatin structure analysis was used and we found that the intensity of sperm DNA damage in DEHP group was significantly increased. Meanwhile, sperm DNA damage in CUR+DEHP group was not significantly affected compared with control group. Moreover, diploid phase rate of testicular cells was significant increased and the percentage of round spermatids was significant decreased in DEHP group compared with control group, respectively. Conclusion: This study provides suggestive evidence of the protective role of CUR on DEHP-induced oxidative stress and sperm chromatin DNA damage. none 許昺奇 2016 學位論文 ; thesis 89 zh-TW
