Summary: | 碩士 === 國立高雄海洋科技大學 === 水產食品科學研究所 === 104 === In this study, fresh water samples were collected from Puzi river in Chiayi County, the fish farm in Jiali District in Tainan city, the spring in Shitan Township in Miaoli County, the river of Maolin in Maolin District in Kaohsiung City, Wanan river in Taiwu Township in Pingtung County, the river of Linrong in Fenglin Township in Hualien County, and seawater sample was collected from stony coral Euphyllia grown in Kenting coast in Pingtung Country. We isolated and identified 147 strains including 140 Gram-negative bacteria and 7 Gram-positive bacteria from these water samples. Phylogenetic analyses based on 16S rRNA gene sequences indicated that strains MI-28, SKY-11, Eup-16, STM-14, STM-24, MVW-6, MVW-40 might be the novel genus and novel species and were subjected to further identification and classification.
Strains MI-28T and SKY-11 were respectively isolated from Puzi river in Chiayi County and the fish farm in Jiali District., Tainan city. Cells are Gram positive curved rod bacteria. Colonies were pale pink coloured, circular, convex and smooth with entire margins. Phylogenetic analyses based on 16S rRNA gene sequences showed that strains MI-28T and SKY-11 form a distinct lineage with respect to closely related genera within the family Microbacteriaceae of the class Actinobacteria, most closely related to Rhodoluna and Pontimonas, and the levels of 16S rRNA gene sequence similarity with respect to the type species of related genera are less than 95%. On the basis of the genotypic, chemotaxonomic and phenotypic data, strains MI-28T and SKY-11 represent a novel genus and species of the family Microbacteriaceae, for which the name Puzihella rosea gen. nov. sp. nov. is proposed. The type strain of the type species is MI-28T (=BCRC 80688T =LMG 27848T=KCTC 29239T).
Strain Eup-16T was isolated from stony coral Euphyllia grown in Kenting coast, Pingtung Country. Cells were Gram negative rod bacteria. Colonies were pale yellow coloured, circular, convex and smooth with entire margins. Phylogenetic analyses based on 16S rRNA gene sequences showed that the closest neighbor strain was Thalassotalea montiporae CL-22T and the sequence similarity was 98.40%. On the basis of the genotypic, chemotaxonomic and phenotypic data, strain Eup-16 represents a novel species in the genus Thalassotalea, for which the name Thalassotalea euphylliae sp. nov. is proposed. The type strain is Eup-16T (=BCRC 80910T =LMG 29001T=KCTC 42743T).
Strain STM-14T was isolated from the spring in Shitan Township, Miaoli County. Cells were Gram negative rod bacteria. Colonies were cream coloured, circular, convex and smooth with entire margins. Phylogenetic analyses based on 16S rRNA gene sequences showed that the closest neighbor strain was Novosphingobium naphtha D39T and the sequence similarity was 98.28%. On the basis of the genotypic, chemotaxonomic and phenotypic data, strain STM-14T represents a novel species in the genus Novosphingobium, for which the name Novosphingobium fontis sp. nov. is proposed. The type strain is STM-14T (=BCRC 80924T =LMG 29290T=KCTC 42983T).

Phylogenetic analyses based on 16S rRNA gene sequences showed that the closest neighbor strain was Novosphingobium piscinae SLH-16T and the sequence similarity was 98.35%. On the basis of the genotypic, chemotaxonomic and phenotypic data, strain STM-24T represents a novel species in the genus Novosphingobium, for which the name Novosphingobium bradum sp. nov. is proposed. The type strain is STM-24T (=BCRC 80925T =LMG 29291T=KCTC 42984T).
Strain MVW-6T was isolated from the river of Maolin in Maolin District., Kaohsiung City. Cells were Gram negative rod bacteria. Colonies were pale pink coloured, circular, convex and smooth with irregular margins. Phylogenetic analyses based on 16S rRNA gene sequences showed that the closest neighbor strain was Flectobacillus lacus CL-GP79T and the sequence similarity was 98.24%. On the basis of the genotypic, chemotaxonomic and phenotypic data, strain MVW-6T represents a novel species in the genus Flectobacillus, for which the name Flectobacillus pallidus sp. nov. is proposed. The type strain is MVW-6T (=KCTC 33800T).
Strain MVW-40T was isolated from the river of Maolin in Maolin District., Kaohsiung City. Cells were Gram negative rod bacteria. Colonies were light yellow coloured, circular, sunk of middle and smooth with undulate margins. Phylogenetic analyses based on 16S rRNA gene sequences showed that the closest neighbor strain was Cellvibrio mixtus ACM 2601T and the sequence similarity was 97.78%. On the basis of the genotypic, chemotaxonomic and phenotypic data, strain MVW-40T represents a novel species in the genus Cellvibrio, for which the name Cellvibrio fontiphilus sp. nov. is proposed. The type strain is MVW-40T (=KCTC 52237T).
