Comparing with proteins, peptides have relative smaller molecular weights, which gives them the variety of physiological activities, such as antibacterial, antioxidant, antihypertensive, antidiabetic, et al. to promote or protect biological systems. Researches on peptides are boosted during the past decades, and still are one of the most important field in medical and agricultural relative academy and industry. However, the fundamental methodology, including peptide extraction, purification, and identification employed for peptide research is almost unchanged and low-efficiency. The conventional approach for specific functional peptides isolation are based on activity-guided fractionation through solvent extraction, preparation column chromatography, and analytical chromatography. Each fraction or eluted peak is subjected to activity assay, e.g. antioxidant assay. The drawbacks of the approach are large amount of sample needed, time-consuming, complicated operation procedures, high cost, and the most seriously, loss the target activity, e.g. antioxidative activity during the approach. Here, we developed a new HILIC (hydrophilic interaction chromatography) /ECD (electrochemical detection)/OPA (post-column O-phthaldialdehyde derivatized) approach for rapid screening and assaying antioxidative peptides with limit amount of samples.
