Summary: | 碩士 === 國立嘉義大學 === 農藝學系研究所 === 104 === Mutagenic efficiency and efficacy of sodium azide (NaN3) in Solanum tuberosum L. was evaluated in this study. The in vitro cultivated node from three cultivars, Kennebec (P86), Tainung 1 (TNG 1), and Tainung 3 (TNG 3), inoculated on amplification medium for 5, 10, 15 days, respectively, were used as the explants. All explants were treated with 0, 0.25, and 0.5 mM NaN3, for investigating the effect of different plantlet age and NaN3 concentrations on survival rate and late blight (LB) disease tolerance. The results showed that increased NaN3 concentration resulted in decreased survival rate of plantlets, and survival rate of plantlets increased when seedling age increased. After NaN3 treatment, all tested potato cultivars showed increased tolerance to LB infection even that these cultivars have different response to LB. Analysis of genetic uniformity of all survived plantlets by RAPD showed that 16, 23, and 14 polymorphic amplicons were found in P86, TNG 1, and TNG 3, respectively. The mutation rate of M1 lines were 37.6%, 28.7%, and 28.2% in P86, TNG 1, and TNG 3, respectively; on the other hand, the result of mutation rate showed that 30.0% by treated 0.25mM NaN3, 38.3% by treated 0.5mM, it seemed that the mutation rate of NaN3 treated plants gradually enhanced by increasing NaN3 concention. In conclusion, we suggested that 10-days-old plantlets with 0.25 mM NaN3 treatment lead to an efficient chemical mutagenesis in potato.
