Sm.) Copel. 6-8 wk after culturing on MS medium supplemented with α-naphthalene acetic acid (NAA). The proliferation of GGBs were stable when they were cultured on MS medium supplemented with the combination of NAA and 6-benzylaminopurine (BAP). Sporophyte regeneration from GGB was classified into three culture stages (early, intermediate and late stages; with a total period of 24 wk). In early stage, GGBs were separated into quarterly excised pellet (QEP GGB) and inoculated on PGR-free 1/2 MS medium. After 12-wk incubation, an approximate production of 851 regenerated sporophytes were obtained from initially cultured 0.1 g young GGB. These regenerated sporophytes went through intermediate and late stage of culture for further development and selection. The qualified plantlets with suitable size were thereafter ready for acclimatization. During the 9-wk acclimatization, sphagnum moss was determined to be the best medium that performed 80% plantlet survival rate. This study also demonstrated that the addition of activated charcoal (AC) into the regeneration medium in early stage significantly reduced browning problems and maintained high sporophyte regeneration from long-term (~ 4 yr) cultured GGB. Based on the optimal culture conditions applied to this study, this in vitro propagation system obtained 284 young plantlets of C. lepifera via culturing 0.1 g GGB for 33 wk. 廖宇賡 學位論文 ; thesis zh-TW
