Part 1: The samples were collected form Q. aliena mycorrhiza in Xinfeng township, Hsinchu county. Part 2: The sample was collected from basidiocarps which grew beneath the Fagaceae species woods in Shekou area, Chiayi county. And the last part, Part 3: The strain sample was isolated and had been proved as a dark septate fungal endophytes by our laboratory. Part 1: There were four endophytes isolated form Q. aliena mycorrhiza (Qa5, Qa8, Qa9 and Qa14). There was no further research on Qa8 and Qa14 strains because they caused death to the host after inoculated. Other strains were compared the database between rDNA sequences and NCBI, the results show that the Qa5 strain belong to Pestalotiopsis sp. and Qa9 belongs to Uncultured ectomycorrhizal. The results of strains characteristic test showed that both Qa5 and Qa9 have the best growth rate in 2% MEA and were capable to secrete peroxide enzymes and protein enzymes. However, only the Qa9 strain had the ability to synthesize IAA. In the synthetic test, only the structure of like-Harting net was observed by Scanning Electron Microscope after inoculating strains on seedlings for 3 months. No mantle had been observed. Part 2: Cms01, which isolated from basidiocarps were found clamp connection in mycelial by micro culture method.It had the highest similarity of rDNA sequences with Pisolithus orientalis (NR119745). The results of strains characteristic test showed the best growth rate for Cms01 would appear in culturing MMN and OFM medium, it could secrete peroxide enzymes and tyrosinases and had the ability to synthesize IAA, which proves Cms01 could accrete with Q. aliena after inoculated for 3 months. Through the observation by Scanning Electron Microscope,we can found observe the characteristics of ectomycorrhizal on mycorrhiza such as obvious mantle and Harting net. Part 3: The dark septate endophytes which isolated and purified by our laboratory was named CkDB2. Mycelial for CkDB2 was dark, septate, and we can observe structure of black chlamydospore by culturing with micro culture method. The results of strains characteristic test showed the best growth rate would happen when cultured with OFM medium, and was capable to secrete tyrosinase and protein enzymes. Besides, it had the ability to synthesize IAA. After inoculating for 3 months, microsclerotia were observed in the mycorrhiza by staining on the root, which proves CkDB2 could with Q. aliena. Sum up with above results, four species of fungus were chosen to go through the compatible tests.The Qa5 and Qa9 strains didn’t form a complete mycorrhiza, the principal reason for this situation could be lack of inoculated time. However, both of them could secrete the enzyme of outside spore, in addition, the Qa9 strain even synthesized IAA. It indicates they have the potential to be the beneficial fungi to plants. The Cms01 strain isolated in this study was confirmed to be P. orientalis, which is new recorded species of Taiwan and was able to form ectomycorrhizal with Q. aliena. In the last part, we observed that the CkDB2 strain could generate dark septate mycorrhiza endophytes with Q. aliena.
